About

The mission of the Single Cell Biomarkers UTechS (scBiomarkers UTechS) is to facilitate biomedical research through high-tech equipment for functional cell assays, multi-color phenotyping, cell sorting, profiling of proteins and transcripts, and single cell multi-OMICS. One of the scBiomarker’s strengths is its L2/L3 laboratory for handling human and infectious material. This configuration and the diversity of its technologies ensure support tailored to all the needs of a biomedical project.

Our support covers all steps of the project, spanning from experimental design, through sample processing, to data analysis. Our current technological portfolio includes 25 cutting-edge technologies installed in 6 dedicated laboratories. Our expertise covers:

  • Cell culture and functional cell assays under controlled gas composition (in Bsl2+ environment)
  • Cell phenotyping and sorting
  • Multiplex and ultrasensitive protein quantification (immunoassays)
  • RNA/DNA profiling in bulk
  • single cell multi-OMICS
  • Data analysis (sc RNA-Seq, multidimensional flow cytometry)
  • Advanced immunomonitoring

The equipment is installed in six dedicated laboratories, enabling optimal experimental setup and sample analysis. A significant part of the equipment is situated within two fully equipped Bsl2+ cell culture laboratories allowing manipulation of human and infectious material.

Since 2011 the scBiomarkers UTechS is ISO9001 certified, providing quality assurance of our service. We are an IBISA-labeled platform.

scBiomarkers UTechS is highly collaborative platform, both nationally and internationally. We are a long-standing partner of Milieu Interieur Project, as well an active member and Center of Excellence of the Federation of Clinical Immunology laboratories (FOCIS).

The scBiomarkers UTechS ongoing developments focus on various novel pipelines for single cell multi-OMICS analysis, and on studies of immune profiles for the discovery of biomarkers in vaccinology and in different pathological contexts.

The scBiomarkers UTechS is affiliated with the Center for Technological Resources and Research (C2RT) and to the Immunology Department of the Institut Pasteur.

HOW TO ACCESS: The scBiomarkers UTechS an open-access facility that welcomes projects and teams from the Institut Pasteur, from the International Network of the Institut Pasteur, from academia, hospitals and industry. It hosts a large number of fundamental research projects, and those with various biomedical topics: infectious diseases, cancer, allergy, autoimmunity, transplantation, etc.

Research groups wishing to include our support in grant applications, or who require help with writing are encouraged to contact us prior to submission.

CB UTechS is an integrated technology platform for multidimensional cell phenotyping & molecular profiling. We propose single-cell cutting-edge technologies and data analysis pipelines. All technologies are bookable through the PPMS online equipment and training reservation site. The equipment is installed in six dedicated laboratories, with a significant part of the technologies situated within BSL2+ (biosafety level 2 +) environment allowing processing of human and infectious material. Cell phenotyping and sorting (Flow Cytometry Platform)

  •  3 Conventional cytometers (Symphony A5, LSRFortessa, Cytoflex for simultaneous detection of up to 28 parameters
  •  Spectral cytometer (ID7000) for indept phenotyping (>40 parameters) and management of autofluorescence
  •  Imaging cytometer MARKII (Luminex): spatial quantification of intra- and inter-cellular processes in cells in suspension
  •  7 Cell sorters (2 Astrios, 4 FACSAria Fusion, 1 FACSAria S6): simultaneous sorting of ≤ 6 cell populations, using ≤28 fluorophores in a single panel. 2 sorters  available in Bsl2+ environment for sorting of human and/or infectious material.
  •  2 Magnetic-based cell sorters (AutoMACS): pre-enrichment of rare cell populations.
  •  MultiMACS: isolation of targeted cell populations from whole blood.

Cell culture and functional assays in Bsl2+ environment (Biomarkers Platform)

  • Fully-equipped cell culture facility
  • Hypoxic cell incubator  XVivo System (Biospherix): manipulation and cell culture under controlled O2, CO2, temperature and humidity
  • Seahorse XFe96 and Seahorse XF Mini Analyzers (Agilent): measuring cellular glycolysis and mitochondrial respiration under controlled O2, CO2, N2, temperature and humidity
  • Incucyte SX5 (Sartorius): short- and long-term live-content imaging in any conventional cell culture support
  • Amaxa nucleofector (Lonza): nucleofection

Multiplex and ultrasensitive protein quantification (immunoassays) (Biomarkers Platform)

  • Bioplex 200 (Biorad) coupled to DropArray (Curiox): quantification of ≤ 200 analytes in cell lysis, supernatants or body-fluids with pM sensitivity from reduced sample (5uL) and reagent volumes.
  • Simoa HD-x (Quanterix): digital ELISA for quantification of proteins with fM sensitivity
  • SP-X (Quanterix): ultrasensitive multiplex protein detection (10-plex)
  • Isolight  (Isoplexis): single -cell sectretome analysis (32-plex)

RNA/DNA profiling in bulk (Single Cell Platform)

  • Nanostring (Nanostring): direct multiplex RNA quantification (800-plex)
  • QIAcuity One 5-plex: nanoplate-based digital PCR application
  • Bioanalyzer & Tapestation 4250 (Agilent): quality control of DNA, RNA, & proteins

Multi-modal single cell profiling (Single Cell Platform)

  • 10xGenomics (3 instruments): single-cell transcriptomics, single-nuclei transcriptomics CITE-Seq, ATAC-Seq, TCR/BCR rearrangement. Available in Bsl2+ confinement for analysis of human and/or infectious cells.
