About

Cytokinesis leads to the physical separation of daughter cells and concludes cell division. Final abscission occurs close to the midbody (or Flemming body), a prominent structure that matures at the center of the intercellular bridge connecting the two daughter cells and first described by Walther Flemming in 1891. The scission occurs not at the midbody itself, but at the abscission site located at distance on one side of the midbody. The first scission is usually followed by a second cleavage on the other side of the midbody, leaving a free MidBody Remnant (MBR). Then, MBRs are either released or wander, tethered at the cell surface for several hours, before being engulfed and degraded by lysosomes. Here, we set up an original method, using flow cytometry, for purifying intact post-cytokinetic MBRs, and identified proteins present – the Total Flemmingsome- and enriched -Enriched Flemmingsome- in this organelle by proteomics.

The Total Flemmingsome is a list of the 1732 proteins identified with at least one unique peptide in EDTA-detached Midbody Remnants (MBRs) purified by flow cytometry (GFP-positive) from HeLa GFP-MKLP2 cells.

The Enriched Flemmingsome is a subset of 489 proteins that were found statistically more abundant in this fraction as compared with at least one of the other controls, the MBR- (GFP-Negative fraction), MBRE (Midbody Remnant-Enriched fraction) or Total cell lysate.

Mammalian proteins composing the Enriched Flemmingsome were individually browsed with their Protein and Gene names for (cytokinesis OR cytokinetic OR midbody OR intercellular bridge OR stembody OR Flemming body OR acto-myosin ring OR actin ring OR central spindle) on the PubMed website. Proteins with a Function in cytokinesis (Role in Cytokinesis) and/or the localization of the protein at the furrow or the intercellular bridge or the midbody (Localized at Furrow/Bridge/Midbody) in mammalian cells are written in magenta (with a green background) and the reference indicated (). Corresponding data can be found in Addi et al, Nat. Commun. 2020 PMID: 32321914 (TableS1-TAB1 and -TAB2)) and are regularly updated. An asterisk (*) indicates when the corresponding reference is based on indirect evidence.
