Cocaine addiction (CocAdd) is a major public health issue affecting 3% of the worlwide population, without validated pharmacotherapy. Nicotinic acetylcholine receptors (nAChRs)have been repeatedly associated in Human and rodent response to cocaine. nAChRs alpha5 subunit locates in key regions for addiction in the brain, with a genetic variant associated with a lower prevalence of CocAdd in three independent case-control studies. Since our collaboration started in 2014, we have gathered further supporting evidence for the links between nAChR alpha5 subunit and CocAdd (Human genetics, self-administration in transgenic rats, in vitro nAChRs concatemers). We now want to further characterize this subunit’s function in CocAdd using complementary, cutting-edge expertise to investigate all its genetic variants by NGS in Human CocAdd, testing the most impactful variants in rodent and molecular models. This will provide a benchmark for biomarkers and pharmacological development in CocAdd.

MECANICOC

La dépendance à la cocaïne (CocAdd) est un problème de santé publique majeur qui touche 3 % de la population mondiale, sans pharmacothérapie validée. Les récepteurs nicotiniques de l’acétylcholine (nAChRs) ont été associés à plusieurs reprises à la réponse de l’homme et des rongeurs à la cocaïne. La sous-unité alpha5 des nAChRs se situe dans des régions clés pour la dépendance dans le cerveau, avec une variante génétique associée à une prévalence plus faible de CocAdd dans trois études cas-témoins indépendantes. Depuis le début de notre collaboration en 2014, nous avons recueilli d’autres preuves des liens entre la sous-unité alpha5 du nAChR et le CocAdd (génétique humaine, auto-administration chez des rats transgéniques, concatémères in vitro du nAChR). Nous voulons maintenant caractériser davantage la fonction de cette sous-unité dans CocAdd en utilisant une expertise complémentaire de pointe pour étudier toutes ses variantes génétiques par NGS dans CocAdd humain, en testant les variantes les plus impactantes dans des modèles de rongeurs et des modèles moléculaires. Cela permettra d’établir une référence pour les biomarqueurs et le développement pharmacologique dans CocAdd.
