Covalent cross-linking combined to mass spectrometry is a powerful approach to address both the structure and composition of protein complexes. To improve the crucial stage of detection and characterization of cross-linked peptides, we have developed a new generation of cross-linking agents based on click-chemistry. The synthesis of these reagent is performed by the group of Prof. Pierre-Yves Renard (University of Rouen).
A new pipeline called e-XL-MS for enhanced-Cross-Linking Mass Spectrometry has been developed recently by our group and published in Analytical Chemistry.
This pipeline is currently extended to in vivo analysis for the analysis of endogenous bacterial protein complexes.
