Abstract:
Aging is the inevitable fate of all organisms yet is highly heterogeneous in nature. It is thought that genetic differences in resistance to environmental stress is one source of individual differences in life a nd healthspan. We have examined the complex genetic basis of natural variation in stress resistance in nematodes using experimental evolution and high throughput functional genomics. Additionally, we have been systematically investigating the influence of genetic background on the response to chemical interventions aimed at extending lifespan. Overall, natural variation for longevity and stress resistance is complex and appears to involve a large fraction of the genome. The large degree of experimental and genetic heterogeneity of these effects suggests that even more precise approaches are needed in our studies. To that end, we are developing microfluidic and single-cell approaches to precisely study the phenotypic and genomic responses to stress and their consequences for healthy aging.
