Fitting tools for integrative structural biology
Modeling cryo-electron microscopy (cryo-EM) reconstructions with computational tools currently enables the interpretation at near-atomic resolution of different functional states of macromolecules, thereby deciphering the functional mechanism of biologically relevant complexes. I will briefly describe the computational algorithms developed by us to build macromolecular models from the information encoded into cryo-EM density maps (see  http://chaconlab.org/methods-bioinformatics-tools). Then, examples of rigid-body or flexible fitting of the available atomic models into selected experimental maps will demonstrate the power of our methodology. These examples include the reconstruction of NAIP5-NLRC4 inflammasome complex and its activation by the bacterial flagellin.
