Title of the seminar : “Expeditions in Genotype-phenotype mapping”

“We can routinely sequence genomes, but now we urgently need tools to interpret individual genomes — multi-level maps that can predict phenotype from genotype. My talk will explore genotype-phenotype mapping at three levels: 1) Towards first-level maps that identify which variants impact gene function, I will describe a deep mutational scanning framework to assess impact of all possible missense variants over the full length of a target protein. Towards second-level maps, capturing the physical network of interacting biomolecules that mediate gene functions, I will describe the Barcode Fusion Genetics Yeast Two-Hybrid (BFG-Y2H) technology to map protein interactions under multiple environmental conditions.  Towards third-level maps that model genetic interactions, the unexpected effect of variant combinations, I will describe use of engineered strain populations to reveal higher-order genetic interactions amongst ABC transporters in yeast.”
