In this project, we build on our previous work of T lymphocyte homeostasis after allogeneic hematopoietic stem cell transplantation (HSCT). We explore early lymphocyte reconstitution and immunological mechanisms of GVHD versus functional immune tolerance.
The main goal of this project is on the one hand to improve our understanding of the cellular and molecular mechanisms at play in the reconstutition of a functional immune system after HSCT, and on the other hand to define biomarkers that can be used in the clinic to predict GVHD development and therapeutic response. To this end, we perform a longitudinal analysis of the phenotypic, molecular and functional characteristics of immune cell reconstitution in patients after HSCT and of the immune cells in their donors, with the goal to define immune parameters of GVHD-related mortality, and of long-term immune tolerance in long-term survivors of HSCT.
