To investigate the molecular processes involved in the synthesis, processing, and trafficking of viral mRNAs and genomic RNAs, we have developed original methods based on engineered viruses expressing a FluPol subunit protein tagged with an affinity tag to perform affinity purification experiments (Fournier et al, PLoS Path 2014), tagged with a split-luciferase domain to detect binary interactions with cellular factors during the course of infection (Munier et al, Mol Cell Proteomics 2013), tagged with a split-GFP domain to document the dynamics of vRNP transport in live infected cells (Avilov et al, J Virol 2012), or tagged with metallothionein to document the ultrastructural features associated with the trafficking of viral genomic RNAs (de Castro Martin et al, Nat Commun 2017).
