A gene targeting vector is transfected into the desired mouse ES cells to modify an endogenous gene by homologous recombination. The mutant ES cells thus obtained are subsequently microinjected into mouse blastocysts for the generation of germ-line chimeras. Together with “classical” 129-derived ES cell lines (i.e.129Sv, 129 Ola, AB1, TC1, CK35Rosa ERT2) we perform currently gene targeting projects using mainly C57BL/6-derived ES cells (C57BL/6N, C57BL/6J).
