About

Le projet ROXANE comporte 3 volets :

1) un volet génétique qui aura pour but d’identifier de nouveaux gènes de l’HS par deux approches

– la recherche au sein de cette cohorte de mutations dans les gènes connus ou les gènes candidats identifiés lors de nos précédents travaux génétiques à l’aide du panel NGS ciblé que nous avons développé

– la poursuite du séquençage d’exome, en s’intéressant à des endophénotypes, des trios sporadiques et de grandes familles multiplexes

2) Un volet de génétique fonctionnelle qui aura pour but d’identifier les voies de signalisation impliquées dans la physiopathologie de l’HS par une approche de transcriptomique

– au niveau tissulaire, comparant la peau lésée de patients HS, la peau péri-lésionnelle de ces mêmes patients et la peau saine de sujets contrôles

– au niveau des kératinocytes, cellules sentinelles de la barrière cutanée, constituant à la fois une barrière physique et immuno-modulatrice, en réponse à des ligands bactériens

3) un volet microbiologique qui aura pour objectif

1- de caractériser le transcriptome des bactéries associées aux lésions d’HS

2-  d’identifier par culture et métagénomique bactérienne les souches bactériennes responsables des résistances aux stratégies antibiotiques

3- de caractériser le microbiote digestif des patients atteints d’HS
