Replication of genomic RNA in viral factories leads to production of viral RNPs (genomic RNA encapsidated by N and associated with polymerase complex proteins). These RNPs have to gain the assembly/budding sites at or near the plasma membrane. Since RNPs are voluminous, their trafficking is expected to rely on cellular machinery. Using recombinant RSV expressing a fluorescent RNP, we set up a pipeline to visualize and quantify RNP movements in living cells. We identify cellular pathway involved in RNPs trafficking. We are dissecting the cellular pathways to identify the proteins and interactions involved in RNP export.
