A CONFERENCE ORGANIZED BY THE SCIENTIFIC COUNCIL

The acquisition of a specific shape is key for organ function. With advances in molecular biology and microscopy, gene networks involved in morphogenesis have been reconstituted and cell behaviour can be traced. A challenge now is to understand how cell behaviour is coordinated to generate shape changes in 3 dimensions. This is the question addressed by the Imagine-Institut Pasteur group of Heart Morphogenesis. We use the developing mouse heart as a striking model of morphogenesis, in which the size and alignment of cardiac chambers are crucial for the functions of contraction and separation of the blood circulations respectively. We are interested in the control of myocardial growth, both in rate and orientation. We have recently identified a novel regulatory pathway, involving the atypical cadherin Fat4, which modulates non-canonical Hippo signalling and restricts heart growth at birth. Another aim is to dissect how left-right patterning drives the asymmetric morphogenesis of the heart. We have recently developed a predictive computer model of asymmetric morphogenesis, from 3D reconstructions of the process of heart looping. Our work in the mouse is relevant to heart repair and congenital heart defects in humans.
