“Clathrin plaques and associated dynamin 2 organize a mechanosensitive compartment”

Abstract :
Large flat clathrin lattices, clathrin plaques, are stable features of the plasma membrane observed in several cell types. They have been associated with sites of strong adhesion suggesting that they could also play a role in force transduction. Here, I will present our work on a compartment composed of clathrin plaques and associated branched actin and demonstrate its requirement for the formation of a three-dimensional intermediate filament web in humans and mice. Importantly, clathrin plaques and associated actin also act as signalling platforms for mechanical stress-induced YAP activity. We show that dynamin 2 participates in clathrin plaque articulation with branched actin filaments and that centronuclear myopathy mutations cause severe plaque and cytoskeleton disorganization and defective mechanical sensing. Together, our data suggest that clathrin plaques, dynamin 2 and associated actin act as platforms for both mechanical signal sensing and transduction which could play an important role in the connection between nuclei and the extracellular matrix.
