In this article, we first present the developmental (lymphocyte differentiation) and epigenetic (locus accessibility
and substrate selection) aspects of V(D)J recombination regulation. We then present recent and earlier work that has shed light on the molecular mechanisms of RAG-mediated V(D)J recombination, with an emphasis on the intricate roles of the recombinase (RAG1 and RAG2), DNA damage response and DNA repair proteins. Finally, we discuss V(D)J recombination in the context of genome maintenance and malignancies of the immune system.
