Les proteusines sont une famille de peptides ribosomiques bactériens qui restent largement élargis des métabolites hypothétiques prédits par le génome. Les seuls membres connus sont les cytotoxines de type polythéonamide avec des structures remarquablement complexes en raison de nombreuses modifications post-traductionnelles habituelles (PTM). Les cyanobactéries contiennent un grand nombre de loci de proteusine putatifs avec des ensembles très variables de gènes candidats PTM. En demandant si cette diversité génétique offre un potentiel de découverte chimique et pharmacologique au-delà des composés de type polythéonamide, nous avons caractérisé le landornamide A, le produit du cluster de gènes osp silencieux de Kamptonema sp.PCC 6506. La reconstruction de la voie dans E. coli a révélé un peptide combinant des lanthionines, des résidus d et, en tant que nouveau PTM, deux ornithines introduites par l’enzyme de type arginase OspR. Le Landornamide A a inhibé le virus de la chorioméningite lymphocytaire infectant les cellules du fibrosarcome de souris, représentant l’un des rares composés anti-arénaviraux connus. Les données soutiennent les proteusines en tant que ressource riche d’échafaudages chimiques, de nouvelles enzymes de maturation et de bioactivités.
