Ces dernières années, plusieurs projets scientifiques sont parvenus à modifier des chromosomes bactériens ou eucaryotes à des échelles jusqu’alors inédites. Les équipes du consortium Sc2.0 ont ainsi récemment remplacé l’intégralité d’un chromosome de la levure Saccharomyces cerevisiae par une molécule très similaire mais entièrement redessinée in silico. À terme, le génome entier devrait ainsi être remplacé par un génome « synthétique ». Dans cet article, nous présentons les principes de l’assemblage de néochromosomes, ainsi que les raisons qui ont fait que la levure S. cerevisiae a été et reste l’un des organismes de choix pour la validation et le développement de ces approches.
