Mass Spectrometry Imaging

Mass spectrometry imaging (MSI) is an analytical technique that involves virtually dividing a surface into a grid of pixels. Each pixel is then individually analyzed using a focused source. This source ionizes the material constituting the sample surface through a phenomenon called desorption/ionization. The generated ions are accelerated and transmitted to a mass analyzer, enabling the measurement of their mass-to-charge ratio (m/z). This process results a set of mass spectra associated with their X,Y coordinates. By using data processing tools, the m/z of an ion can be selected to reconstruct a density map (see Figure 1).1 The spatial resolution in MSI depends on the diameter of the probe (focused source). The advantage of MSI is its ability to determine the spatial distribution of various analytes without requiring labeling. This technique has numerous applications, such as metabolomics, lipidomics, pharmacokinetics, etc…2–6

Quentin Vanbellingen, Ph.D.Research Engineer

Core expertise: Mass spectrometry (MALDI, TOF-SIMS), high-resolution molecular imaging, surface analysis, pharmacokinetics, experimental design, data processing and visualization.

Ph.D.CNRS / Université Paris-Saclay (ICSN) (2012-2015): Developed a delayed-extraction TOF-SIMS acquisition mode, enabling the combination of submicrometer spatial resolution with suitable mass resolution for imaging organic compounds in biological and inorganic samples.

Postdoctoral fellow Florida International University (USA) (2016): Investigated intracellular drug absorption and distribution at the subcellular scale using TOF-SIMS imaging; designed and implemented innovative experimental and analytical protocols.

Pharmaceutical industry Technologie Servier (2017-2022): Study director for preclinical and clinical ADME projects; led programs on small molecules and therapeutic oligonucleotides, structural analysis of exogenous metabolites, PK measurements, and regulatory aspects. Head of the biodistribution platform.

Academic research CNRS / LAMS (2022-2023): Applied mass spectrometry to the study and preservation of cultural heritage materials; data processing and multimodal integration.
