Despite effective prevention against hepatitis B virus (HBV )infection, 300 million people worldwide are chronic HBV carriers, of whom 25% will die of liver cirrhosis or hepatocellular carcinoma (HCC). Although antiviral treatments of chronic HBV carriers, including pegylated interferon-alpha and inhibitors of the viral polymerase, control efficiently virus replication and improve liver functions, they cannot lead to a complete viral clearance. This failure is due to the inability of the treatment to eliminate the viral DNA present in the nucleus of infected cells as a covalently closed circular DNA (cccDNA). cccDNA serves as a template for the transcription of all viral RNAs and thus constitutes a viral reservoir responsible for viral rebound at the end of the treatment. Our group investigates mechanisms regulating HBV cccDNA transcription and persistence.
