A SPOC for using the Agilent Bioanalyzer
Training sessions for the use of the Bioanalyzer, organized at the Transcriptome and Epigenome platform of the Biomics pole, are now replaced by a SPOC, available 24 hours a day on-line via the Pasteur Institute’s MOOC platform.
This online privatized course (SPOC) is organized in several modules, so that you become autonomous to use the Bioanalyzer.
At each step, you will be asked to answer a quiz-memory, and, at the end of the training session, you will get your certificate, allowing you to have access to the Bioanalyzer of the platform. Connect yourself !

The Agilent Bioanalyzer
The Bioanalyzer 2100 is dedicated to electrophoresis of DNA, RNA and proteins in a microfluidic circuit. The loading of samples is done on specific chips based on the type of sample. Electrophoresis is fast and provides qualitative data on the samples.
The training on the on-line course (SPOC) is performed on a RNA 6000 Nano Agilent chip, for quality control of the RNA preparations. A small amount of RNA (1μl at 50 – 400 ng) was loaded in each well in the presence of a fluorescent label. The instrument measures in real time the fluorescence intensity emitted as a function of the RNA retention times in the microarray and the concentration of RNA molecules. The retention time and fluorescence intensity data are validated by reference to the molecular weight marker. The results are visualized in the form of an electropherogram from which the software evaluates the integrity of each RNA preparation and assigns a quality index (RNA Integrity Number) to each sample.
The main advantages of using this technology for the analysis of RNA preparations are: the speed of analysis, the small sample and reagent consumption, as well as the standardization and automation of the process.

The Bioanalyzer PF2 is “self-service” under two conditions.
It is essential to book in your name -and exclusively- the desired time slot for your manipulation via the booking system (PPMS,http://www.pasteur.fr/ppms/login/?pf=13 ). People using the system under the name of a colleague must contact us promptly.
Billing for the service is always made via a paper sheet called « Bon de cession interne » (or BCI). Not needed to be completed or signed now, but DO NOT forget to indicate your “CODE d’IMPUTATION BUDGETAIRE” !
