  1. Establishment of inability of HSCs to transdifferentiate into non-hematopoietic lineages. Through detailed in vivo lineage tracing experiments in mouse models of bone marrow transplantation, the Jacobsen lab failed to confirm multiple high-profile studies which had implicatied that HSCs in adult bone marrow could efficiently “transdifferentiate” into non-hematopoietic fates in the injured heart and pancreas. In addition to finding no evidence for the claimed ability of HSC to transdifferentiate upon transplantation but rather at low efficiency resulting in heterotypic cell fusion, the Jacobsen lab demonstrated that transplantation of bone marrow cells into an infarcted heart on the contrary could potentially result in severe side effects. These findings contributed in a decisive manner towards halting attempts to widen the indications of clinical bone marrow transplantation for regeneration in non-hematopoietic tissues.

  1. Revisions of the hematopoietic hierarchy and lineage commitment map. Studies from the Jacobsen lab have been instrumental in unravelling novel lineage commitment pathways, resulting in redrawing of the “textbook” hierarchical map of normal hematopoiesis. This includes the identification and characterization of lymphoid-primed multipotent progenitors (LMPPs), as a key stage in early mouse and human hematopoietic lineage fate decisions and as the first commitment stage at which lymphoid transcriptional priming is observed, implying an early separation of megakakaryocyte-erythroid and lympho-myeloid immune-restricted lineage commitment pathways, rather than the classical model in which a strict separation into a common myeloid and a common lymphoid pathway had been implicated to be the first lineage restriction step of HSCs. The Jacobsen lab also established that the progenitors that seed the embryonic thymus and those replenishing the thymus postnatally have combined myeloid, B and T cell but not megakaryocyte-erythroid potential, and that the first LMPPs emerge early in embryonic development, prior to definitive hematopoietic stem cells. In other studies distinct and early separated lineage commitment pathways for Gata1-dependent and independent myeloid blood cell lineages were established. Of particular relevance for malignant hematopoiesis, the Jacobsen lab also provided evidence in support of LMPPs and ETPs being target cells for transformation in myeloid leukemias and in clinically distinct mixed lineage phenotype ETP leukemias.

  1. Identification of a distinct repertoire of multipotent HSCs adopting lineage-restricted including platelet-restricted fates in vivo.

The Jacobsen lab established that a fraction of hematopoietic stem cells are distinctly transcriptionally primed for the megakaryocyte-platelet lineage, and in support of a separate and early lineage commitment pathway for the megakaryocyte lineage, they identified, prospectively purified and characterized a distinct population of self-renewing stem cells highly biased towards replenishment of the platelet lineage . Later they demonstrated that a distinct class of HSCs adopts a fate exclusively towards effective and stable replenishment of a megakaryocyte/platelet-lineage tree but not of other blood cell lineages, despite of sustaining multipotency, whereas no HSCs contribute exclusively to any other single blood-cell lineage. They also showed that these platelet-restricted HSCs become increasingly dominant in aged mice, and that another distinct class of HSCs can fully restrict towards simultaneous replenishment of megakaryocyte, erythroid and myeloid lineages without executing their sustained lymphoid lineage potential. Genetic lineage-tracing analysis also provided evidence for an important role of platelet-biased HSCs in unperturbed adult haematopoiesis. These findings uncovered for the first time a limited repertoire of distinct HSC subsets, defined by a predictable and hierarchical propensity to adopt a fate towards replenishment of a restricted set of blood lineages, before loss of self-renewal and multipotency.

  1. Identification and characterization of cancer stem cells in MDS, myeloproliferative neoplasms and childhood leukemia, including their ability to selective evade otherwise effective therapies.

The Jacobsen lab prospectively identified and characterised rare Myelodysplastic Syndromes (MDS) stem cells, establishing their origin in the normal hematopoietic stem cell compartment, and that in early stages of MDS, sequential acquisition of recurrent oncogenic mutations occurs exclusively in this MDS stem cell compartment, demonstrating that these mutations while giving a clonal advantage at the stem cell level are insufficient to introduce self-renewal properties in hematopoietic progenitors, establishing that these distinct and rare MDS stem cells are both sufficient and required to propagate the malignant clones in early stages of MDS. These were the first studies, which established the identity of human cancer stem cells independently of using controversial in vivo xenotransplantation assays demonstrated to fail to reliably uncover cancer stem cell activity. Moreover they also demonstrated that del(5q) MDS stem cells selectively escape Lenalidomide treatment in patients reported to be in complete remission. In other studies, the Jacobsen lab identified and characterized the malignant stem cells in childhood leukemia and myeloproliferative syndromes.

An international review panel (Oxford evaluation Panel) which evaluated all research programs at the Medical Faculty, Lund University, in 2002, ranked the Stem Cell Program as ”the clearly strongest and most coherent research program at the Medical Faculty, Lund University”.

2003-2008: Founding Director of Lund Stem Cell Center (SCC). Initiative and coordinator for successful application to Swedish Foundation for Strategic Research (SSF) for “Future Research Leaders” (SEK 100 milion awarded to SEJ as PI) to become one of the first Swedish Centers of Excellence in Life Sciences, the Lund Strategic Research Center for Stem Cell Biology and Cell Therapy, a translational research center focussed on basic and translational stem cell and developmental biology of the blood and central nervous system, HSC transplantation and a world leading clinical transplantation program for Parkinson’s Disease. Raised new funding in excess of 20 million Euro to develop the center, led successful international recruitment efforts of six young clinical scientist group leaders in the fields of adult and paediatric HSC transplantation as well as immune and gene therapy and cancer stem cell biology. Through this process he has also raised considerable funding for these positions and the corresponding translational research projects, as well as infrastructure support and technology platforms (including GMP facilities) facilitating clinical stem cell research at the Lund SCC. Notably, when the Lund SCC, one of six Swedish Centers of Excellence (CoE) in life sciences, underwent a mid-term (three year) international evaluation in 2005, the SCC was in particular commended for their successful efforts in strengthening the translational research and strong international recruitments. As a consequence of this the Lund SCC was lifted from a sixth to third ranking of the Swedish CoEs, and as a consequence of the strong development the international review panel recommended the SCC to receive the highest level of additional funding of the CoEs. The Lund SCC remains an internationally leading research center for basic and translational stem cell research.

2006: First Professor and Chair in Stem Cell Biology and Regenerative Medicine at University of Oxford, and establishment of Laboratory of Haematopoietic Stem Cell Biology at Weatherall Institute of Molecular Medicine.
