Muriel Gugger’s lab – Pasteur Culture collection of Cyanobacteria (PCC)
“Cyanobacterial bioactive compounds” project

Cyanobacteria are important primary producers and promising producers of natural products. Secondary metabolism of cyanobacteria represents already over 1100 secondary metabolites reported from 39 cyanobacterial genera. With the development of biotechnological tools and genomic data analysing, there is an increasing interest in using cyanobacteria as cell factories for the production of biomolecules. Notably antimicrobials from cyanobacteria are of particular interest in the frame of challenge and new concept in antibiotic research.
Genome mining of several cyanobacteria revealed uncharacterized antimicrobial molecules encoded by highly conserved and compact biosynthetic gene clusters. Using molecular biology (heterologous expression and CRISPR), we will study the rational behind the generating diversity of antimicrobial molecules. This will allow us to further test the antimicrobial activity of each produced antimicrobial molecules.

PlastiCure is a biological system designed to degrade polyethylene terephthalate (PET) and use the degradation products to synthesize the erythromycin A. For this, a chain able to degrade PET into propionyl-CoA will be inserted in an Escherichia coli strain. This molecule will be the precursor of the synthesis of the erythromycin A, a commonly used antibiotic. 

The objective was to assemble the genes coding for the enzymes of the PET degradation pathway in one operon. For this, we chose the Gibson Assembly, a novel method that allows to assemble until 6 DNA fragments. Whereas, our operon is composed by 13 fragments of 15 genes. We decided to divise the assembly into 5 gene clusters composed by 2 to 4 genes of the degradation pathway. Then this 5 clusters will be purified and assembled to form the operon of the PET degradation pathway.

Internship in TaMaRa’s lab – U1001 (ex U571) from INSERM.
Teaching and promotion of science by the creation of learning kits for synthetic biology, intended for secondary school students and public. This project proposes a series of didactic biology kits that allow the user to get familiar with basic concepts of microbiology and molecular biology. It has been created by the Open Science School , from the Center for Research and Interdisciplinarity (CRI).
– Did experimental works in microbiology on BSL 1 strains,
– Wrote documentation of the strains (booklet),
– Participated to educational activities with high school students in the laboratory.
