2023-2024, 2d semester

Colorectal cancer (CRC) is the second cause of cancer-related deaths in France and abroad. Among its multifactorial causes are dysbiotic microbiota and the chronic exposure to resident pathobiont, such as pathogenic strains of Escherichia coli.

Our laboratory works in the Pasteur Institute on pathogenic strains of E. coli which have the capacity to produce toxins that manipulate host cell signaling pathways and generate genomic damages.

The aim of the project is to address the pro-oncogenic role of the CNF1 toxin in a mouse organoid model, specifically measuring impact of the toxin on their growth potential, epithelial inflammation, proliferation, tumor-specific signaling and DNA damage by whole mount immunofluorescence, western-blot, ELISA and gene expression analysis. Organoids will be derived either from WT mice or from knockouts in genes of susceptibility to cancer.
