Innate lymphoid cells (ILCs), including natural killer cells, are a diverse family of hematopoietic effectors that rapidly produce cytokines and can play anti- or pro-inflammatory roles in infection, transformation and tissue homeostasis. The roles for NK cells in the response to tumors or microbial infections are well described explaining the steadily increasing number of immunotherapies targeting NK cells. However, the impact of other ILC subsets in pathophysiologic conditions remains less defined.
Based on a large screen of NK cells and ILC in a novel genetic mapping mouse resource, called the Collaborative Cross (CC), we identified through quantitative trait locus mapping candidate genes driving three key NK cell traits and further genomic loci involved in the regulation of 24 different NK/ILC traits. Moreover, we identified a selection of CC strains that represent novel mouse models to study NK cell/ILC differentiation and anti-tumor functions. This project aims at confirming candidate genes that influence NK cell homeostasis and activity as well as ILC differentiation.
This project involves state-of-the art molecular and cellular methodology as well as bioinformatical analyses.
